Agree that one of the main issues with removing labels is when it is free text entry. As in drop downs/select boxes the user can still see the placeholder text, if it is includes as the default option. Not sure if this would have a negative impact on accessibility though?

I'd agree with contrast over colour. I'd hypothesise that smaller details would be the first to be obscured by bright lighting. From personal experience I have used both light on dark and dark on light (foreground / Background) - I didn't experience any discernible difference.

You talk of familiarity decreasing? How is this been measured? Are less users are clicking the button the longer they use the system? Without further details its hard to provide an answer. Think about the task they are trying to complete and what the button does.

I'd agree. For example: Each post on a forum I want to see latest (or latest with activity) at the top. But in the comments section I want to start with the oldest so I can follow the conversation.
